Samuel Millen was born on Easter Sunday in 1818. He was educated at a Chester District public school. He lived in South Carolina and Louisville, Kentucky, before moving to Bloomington to attend college in 1838. He graduated from IU in 1842. His lifelong occupation was as a minister of the Gospel in the United Presbyterian Church. He held the position of pastor for the Shiloh Congregation in Indiana for three years, in Clayton, Indiana, for seven years, and for Smith Creek in Illinois for fifteen years. He was prominent in the movement to establish Illinois’ Monmouth College in the 1850s and was connected with the U.S. Christian Commission during the Civil War.
In 1846, Millen married Mary Wier. His namesake son, Samuel Rutherford Millen, was a medical doctor; his grandson, William Millen, was a minister in the Indian Territory (now Oklahoma).
